Tryptic Soy Broth (TSB) is a general purpose culture broth widely used in Antimicrobial Textile Testing to support the growth of microorganisms.
TSB PREPARATION
TSB can be purchased as a powder that has to be transformed into a solution before its use. Typically, TSB solutions are used as liquid media to prepare the Single Colony Growth, as one of the components of the Suspension Medium, and also as a medium to perform serial dilutions.
These are the instructions to prepare 100 ml of TSB using Thermo Scientific™ Remel™ Tryptic Soy Broth:
INSTRUCTIONS
- Measure 100 ml of deionized water using a graduated cylinder
- Place the stir bar in a flask
- Weight 3 gr of TSB powder using an analytical balance and add it to the flask
- Add the water
- Place the flask onto the stir plate and stir and warm for at least 2 minutes or until the visible clumps break down
- Transfer the obtained solution into a media bottle, close the lid loosely, label, and put autoclave tape
- Autoclave for 15 minutes at 121 ºC
MATERIALS
- 3 gr of TSB powder
- 100 ml of deionized water
- Magnetic stir plate and stir bar
- 250 ml flask
- 200 ml media bottle

SPECIAL CONSIDERATIONS
- It is not necessary to use sterile deionized water to prepare the TSB, since the solution will get sterilized during the autoclave process.
- If used to prepare Single Colony Growth, during its preparation special attention should be given to selecting a media bottle that will later fit into the Incubator Shaker.
- After preparation, the TSB solution has to be cooled down to room temperature before its use.
- TSB can be stocked in the refrigerator at 4 ºC, and before its use it has to be acclimated to room temperature again.
