TSA Petri dish

Some microbiology techniques like streak plating require the use of TSA dishes. These plates can be either purchased, or prepared in the laboratory. Follow these instructions to prepare TSA petri dishes.

INSTRUCTIONS

  1. Prepare the TSA solution
  2. Cool down to 55 °C
  3. On a sterile petri dish, slowly dispense 15 ml of TSA using a serological pipette, trying to avoid the formation of bubbles
  4. Let cool down inside the hood for 30 minutes with the lid open

MATERIALS

  • 15 ml of TSA sterilized solution
  • 15 ml serological pipette
  • Pipette controller
  • Sterile petri dish

SPECIAL CONSIDERATIONS

  • The plate should not be moved during the cool down process to ensure a uniform surface
  • If not used immediately after preparation, TSA petri dishes can be stored in the following manner:
    • Turn the plates upside down to prevent moisture to condense onto the agar surface.
    • Place the plates on a plastic bag, and store them at 4°C following the TSA manufacturer´s shelf life instructions.
